on a guitar, does it make a difference which way round the pickups are?

as in, if you spun each pickup around so it was the other way up, would it make a difference to the sound? took my guitar apart and dont know if i put the pickups back the same way!

You mean turn the pickups through 180 degrees, so that what was the leftmost pickup is now on the right?

It would only make a difference if your pickups are "tuned" so that the leftmost ones are more sensitive to bass frequencies and the right ones sensitive to higher frequencies.

What guitar do you have? What type of pickups are they? Humbuckers?

usually the wire comes out of one side of the pickup. If you face the pickups so that the wire is coming out of the pickups near the knobs it's usually that way round.

hope that makes sense (i have a hangover)
